A certificate of analysis (COA) for a compound amino acid powder may report total nitrogen, organic nitrogen, amino nitrogen, and individual nitrogen forms such as ammonium or nitrate.

These measurements describe different aspects of the product. They should not be treated as interchangeable, and none of them alone proves biostimulant performance.

The distinction between TN and TKN matters. EPA Method 351.2, for example, defines TKN as ammonia plus organic nitrogen. That water-analysis method illustrates the terminology; it is not, by itself, a recommended method for amino acid powder. Ask for a procedure validated for the product matrix. EPA Method 351.2

Urea needs separate treatment. Chemically, it is an organic compound, but it is not an amino acid or peptide. Do not assume that a result labeled “organic nitrogen” excludes urea. Ask the laboratory or supplier to explain the definition used.

Organic Nitrogen Is Not a Measure of Bioactivity

Organic nitrogen can occur in amino acids, peptides, proteins, and other compounds. The category does not identify which substances are present or how they will perform in a crop application.

Likewise, a higher free amino acid percentage does not automatically make a product more effective. It may indicate differences in formulation or the extent of hydrolysis, but it does not capture the full peptide composition.

Total Amino Acids, Free Amino Acids, and Amino Nitrogen

These three results answer different questions.

Total amino acids generally refers to the amino acids quantified after a defined hydrolysis procedure, including those released from peptides and proteins. Confirm which amino acids the method covers and how the total is calculated.

Free amino acids are measured without first hydrolyzing the peptide-bound fraction. An amino acid profile measured after hydrolysis cannot establish the original free amino acid content.

Amino nitrogen measures an analytical response from amino groups. Depending on the method, free amino acids, peptide terminal groups, and other substances may contribute. It is not a direct substitute for a chromatographic amino acid profile.

HPLC identifies an analytical platform, not a complete method. Sample preparation, separation, calibration, and detection all matter. Hydrolysis can also alter or destroy certain amino acids, so laboratories may need additional procedures for a fuller profile. FAO analytical guidance

How to Check Whether the Numbers Are Consistent

1. Match the Reporting Basis

Before comparing percentages, confirm that they refer to:

For example, a urea percentage cannot be subtracted directly from a TN percentage. It must first be converted to urea nitrogen.

2. Avoid a Universal 0.16 Conversion Factor

Where the individual amino acid concentrations and reporting conventions are known, nitrogen attributable to the measured amino acids can be estimated from their molecular composition:

Estimated amino acid nitrogen = Σ [amino acid concentration × nitrogen mass fraction of that amino acid]

The calculation must account for the chemical form and analytical reporting basis. A laboratory should review it when hydrolysis changes the identities of the compounds being measured.

3. Investigate Unexplained Nitrogen

Consider a hypothetical powder with:

Assuming these are non-overlapping measurements on the same basis, the remaining 5.1 percentage points require explanation.

Possible causes include other nitrogen-containing compounds, incomplete analytical coverage, inconsistent reporting bases, or measurement problems. This discrepancy does not by itself prove adulteration, but it should be resolved before making a quality claim.

Do not automatically attribute the difference to peptides: a total amino acid analysis after hydrolysis should already include the peptide-derived amino acids it successfully recovers.

What Buyers Should Request

Ask the supplier for:

  1. A COA linked to the lot being purchased, with actual results distinguished from specification limits.
  2. The nitrogen method, including any modification used to recover nitrate or other nitrogen forms.
  3. The definition and calculation used for organic nitrogen.
  4. Separate total and free amino acid results, with sample-preparation details.
  5. The individual amino acid profile and the list of analytes included in the total.
  6. Moisture content and the reporting basis for every percentage.
  7. Disclosure of added nitrogen sources and relevant formulation ingredients.
  8. Supporting product-performance data where efficacy claims influence the purchase.

Also check sampling dates, laboratory identity, and whether the sample represents one lot or a composite of several lots.

Frequently Asked Questions

Is higher total nitrogen always better?

No. It means more nitrogen per unit of product on the stated basis. Whether that is desirable depends on the intended formulation and application.

Can I calculate organic nitrogen from the amino acid profile?

Not completely. A profile can help estimate nitrogen attributable to the measured amino acids, but it does not account for every possible organic nitrogen compound.

Does ammonium or urea prove that a product is adulterated?

No. A product may intentionally contain additional nitrogen sources. The important questions are whether the formulation is disclosed and whether it matches the agreed specification.

Does organic nitrogen mean the product is approved for organic farming?

No. Chemical terminology does not establish certification or eligibility for a particular organic production system. Request documentation applicable to the intended market and use.

Can total amino acids minus free amino acids give the peptide content?

With compatible methods and reporting bases, the difference may estimate the amino acids recovered from the bound fraction. It does not directly measure peptide mass, peptide size, or biological activity.

Make the COA Part of a Broader Evaluation

Use total nitrogen to understand nitrogen loading, amino acid analysis to assess composition, and relevant trials to evaluate performance.

Consistent analytical results support a purchasing decision, but they do not replace formulation transparency or evidence for the intended use.
